艾美捷PEPC丨磷酸烯醇丙酮酸羧化酶 应用实例:

10µg从拟南芥wt叶组织(Atn非衰老叶)、拟南芥wt叶组织(Ats衰老叶)、红松针叶组织(PS0、PS36)中用1 M Tris HCl、pH 6.8、10%SDS、15%蔗糖新鲜提取的总蛋白,在10%Bis-Tris Nupage Novex凝胶(120 V/45 min,使用MES缓冲系统)上分离0.5 DTT和在75°C下变性5 min,并在30 min内用聚偏氟乙烯(PVDF)吸附。搅拌45分钟/RT,用5%脱脂牛奶阻断印迹。在含有2%脱脂乳的TBS中搅拌,或在/4°C下搅拌,以1:1000的稀释度在一级抗体中孵育1h/RT。倾析抗体溶液,在RT条件下搅拌,在TBS中短暂冲洗印迹两次,持续10分钟。在Agrisera匹配二级抗体(抗兔IgG-辣根过氧化物酶结合物,AS09 602)中培养印迹,稀释至1:75 000 in,搅拌1h/RT。如上所述清洗印迹,并使用化学发光检测进行显影。曝光时间为40秒。
